This lecture builds directly on Part 1 by showing how the proton motive force generated across the inner mitochondrial membrane is harnessed by ATP synthase (Complex V) to produce the energy currency of the cell. We begin by reviewing the separation of protons into the intermembrane space, creating both a chemical gradient (pH difference) and an electrical gradient (membrane potential). Together, these form the proton motive force, which drives protons back into the mitochondrial matrix through the F₀ portion of ATP synthase. This rotation of F₀ induces conformational changes in the F₁ subunit, catalyzing the formation of ATP from ADP and inorganic phosphate.
